Pyrophosphate-site inhibitor
Viral DNA polymerase inhibitor — pyrophosphate site
Pyrophosphate-site inhibitor — mechanism card
- A pyrophosphate analogue that blocks the pyrophosphate-binding site directly — no viral or host kinase activation
- Works against acyclovir- and ganciclovir-resistant HSV/CMV because it needs no thymidine kinase or UL97
- Nephrotoxicity and electrolyte wasting (Ca, Mg, K, phosphate) → seizures

Drugs with this mechanism (1)
- Foscarnet — Needs no viral kinase, so it works against resistant CMV and HSV.
