GABA/glycine release inhibitor
SNARE-mediated GABA/glycine release inhibitor
GABA/glycine release inhibitor — mechanism card
- Cleaves VAMP/synaptobrevin → inhibitory vesicles cannot fuse and release
- Acts on the spinal inhibitory interneuron, so glycine and GABA stop reaching the motor neuron
- Disinhibition, not weakness: trismus, risus sardonicus, opisthotonus, reflex spasms
- Travels to the cord by retrograde axonal transport; botulinum stays at the neuromuscular junction

Drugs with this mechanism (1)
- Tetanus toxin — Clostridial protease that silences inhibitory interneurons — spastic paralysis, the mirror image of botulinum.
