- Blocks the MUTANT IDH1/IDH2 enzyme, whose neomorphic activity makes 2-hydroxyglutarate
- Falling 2-HG restores normal DNA and histone demethylation, so the tumor differentiates
- Only useful where an IDH mutation has been demonstrated
