Cell-depleting antibody
Antibody-mediated immune-cell depletion
Cell-depleting antibody — mechanism card
- Marks a cell for destruction rather than blocking a signal — the target cell disappears
- The four CD20 antibodies deplete B cells: rituximab, ocrelizumab, ofatumumab, ublituximab
- Benralizumab binds IL-5Rα but is here, not with the receptor blockers, because the point is ADCC depletion of eosinophils
- No toxin payload — the antibody recruits the patient’s own effector cells and complement

Drugs with this mechanism (5)
- Benralizumab — IL-5 receptor antibody that depletes eosinophils; severe eosinophilic asthma and EGPA.
- Ocrelizumab — Anti-CD20 antibody — the first drug approved for primary progressive MS.
- Ofatumumab — Self-injected anti-CD20 antibody for relapsing MS; the same molecule is ARZERRA in CLL.
- Rituximab — Anti-CD20 antibody; screen for hepatitis B before every course.
- Ublituximab — Glycoengineered anti-CD20 antibody for relapsing MS, infused in one hour.
