PCSK9 siRNA
PCSK9 small interfering RNA (siRNA)
- Small interfering RNA that silences hepatic PCSK9 messenger RNA before the protein is ever made
- Less PCSK9 means more LDL receptors recycled to the hepatocyte surface, so LDL-C falls
- GalNAc conjugation targets the hepatocyte; dosing is twice yearly after the loading doses
Drugs with this mechanism (1)
- Inclisiran — Liver-targeted PCSK9 siRNA for durable LDL lowering in ASCVD and familial hypercholesterolemia.
