Protozoal dihydrofolate reductase inhibitor
- Selective for the PROTOZOAL enzyme — Plasmodium, Toxoplasma
- Selectivity is narrower than trimethoprim’s: human marrow is hit at therapeutic doses
- Leucovorin is given alongside to protect the marrow, not to reverse the drug
