Metabolism (active-metabolite dependent)
- Clinical effect depends partly on active metabolite
- Parent metabolism creates pharmacologically active drug
- Enzyme inhibition may reduce or increase net effect
- Parent level alone may not predict response
- Half-life can reflect both parent and metabolite
Interaction at a glance
- Clinical effect depends partly on active metabolite
Mechanism
- Clinical effect depends partly on active metabolite
Management
- Parent metabolism creates pharmacologically active drug
- Enzyme inhibition may reduce or increase net effect
Magnitude / time course
- Parent level alone may not predict response
- Half-life can reflect both parent and metabolite
Self-check
- How may enzyme inhibition affect an active-metabolite-dependent drug?
Reduce or increase net effect.
- Does parent level alone necessarily predict response with an active metabolite?
No.
